MyGlo™ Reagent Reader: A Scientific Advancement in Luminescence-Based Assays

The MyGlo™ Reagent Reader by Promega enables high-sensitivity luminescence-based cell viability assays with cloud-based data management.

The MyGlo™ Reagent Reader, by Promega Corporation, is a state-of-the-art luminescence detection system. This compact, high-sensitivity instrument integrates assay setup, luminescence measurement, and data analysis, offering life science researchers an efficient and reliable tool. The device is optimized for luminescence assays, currently loaded with seamless configuration for analyzing CellTiter-Glo® and Caspase-Glo® 3/7 Assays (in 2D or 3D), providing accurate quantification of cell viability or apoptosis, respectively. This paper outlines the key specifications, performance metrics, and advantages of MyGlo™ over conventional plate readers, solidifying its role in modern biological research.

Luminescence based assays play a pivotal role in life science research, providing crucial insights into cell health, proliferation, and response to chemical agents. Traditional luminescence-based detection methods require large, costly, and complex instrumentation. The MyGlo™ Reagent Reader addresses these challenges by offering a portable, efficient, and cost-effective alternative with a broad dynamic range and cloud-based data management.

    Experimental Design

To evaluate the performance of the MyGlo™ Reagent Reader, a luminescence-based ATP detection assay was conducted using CellTiter-Glo®. The luminescence signal was recorded across multiple cell densities to determine the device’s sensitivity and dynamic range.

The MyGlo™ Reagent Reader demonstrated a strong correlation between cell density and luminescent output, confirming its high sensitivity and precision in detecting ATP levels.

Jurkat cells were seeded in all-white 96-well plates from 10 cells/well to 100,000 cells/well (n=8 replicates). Cell viability was determined by adding an equal volume of CellTiter-Glo® or CellTiter-Glo® 2.0 Reagent to all wells, and the signal was read on a MyGlo™ Reagent Reader. These graphs were generated using Microsoft Excel.

These results highlight the broad detection range and robustness of the MyGlo™ Reagent Reader, making it suitable for various applications in pharmacology, toxicology, and biomedical research.

Compared to conventional multimode plate readers, the MyGlo™ Reagent Reader offers several distinct advantages:

    • Cost Efficiency: Provides high-performance luminescence detection at a lower price point than larger multimode plate readers.
    • Simplified Workflow: Eliminates the need for extensive calibration and complex operational protocols.
    • Cloud-Based Data Management: Enables remote access, storage, and collaborative data analysis, which is lacking in standard microplate readers.

The MyGlo™ Reagent Reader is a highly efficient, precise, and portable solution for luminescence-based assays. Its superior sensitivity, broad dynamic range, and cloud integration position it as a leading alternative to traditional multimode readers. As a cost-effective and easy-to-use tool, MyGlo™ is poised to revolutionize luminescence assessments in both academic and industrial research settings.
